    The Dana-Farber Cancer Institute seeks a research technician to focus on next-generation, translational pathology studies who will work within the Wolpin-Nowak laboratory: ******************************************** as part of the Hale Family Center for Pancreatic Cancer Research: *************************************** and the Lustgarten Foundation Pancreatic Cancer Research Laboratory at the Dana-Farber Cancer Institute. Our research group investigates the biology and risk factors associated with pancreatic and other gastrointestinal cancers. We aim to improve outcomes for patients with these diseases both by identifying cancer earlier, when the chance for a cure is greater, and by improving therapy. We focus on learning from cancer patients by studying human biospecimens so that our discoveries can be rapidly and confidently translated into improved clinical care. Our laboratory members include epidemiologists, biostatisticians, pathologists, data scientists, molecular biologists, and clinical researchers, all of whom contribute their unique expertise to our lab's work. We employ a broad variety of analysis approaches and techniques in our laboratory, including population-scale epidemiology studies, digital pathology, genomic sequencing, and spatial transcriptomic and protein profiling. Our tissue profiling work encompasses both clinical trial correlative studies and landscape-directed discovery studies. We work closely with the Gastrointestinal Cancer Center Biobank to acquire blood, tissue, and other sample types from patients at Dana-Farber Cancer Institute. Finally, we routinely collaborate both with investigators at Dana-Farber and across the nation to study pancreatic cancer using model systems and to conduct and lead clinical trials. May be responsible for ordering supplies. Collaborate with purchasing. The Research Technologist will work independently with appropriate supervision from Dr. Turbett and investigators in the Division of Infectious Diseases, with an emphasis on rigorous documentation, contamination control, quality control (QC), and troubleshooting. All testing performed in this role is for research and/or surveillance purposes (non-clinical reporting); clinical technician certification is not required. Qualifications * Perform and optimize nucleic acid extraction workflows (manual and/or automated) for a variety of human clinical specimen types; implement extraction controls and contamination prevention practices. * Develop, optimize, and troubleshoot high-complexity molecular assays including RT-qPCR, qPCR, and digital PCR. Prepare and interpret run level QC metrics and troubleshoot pre-analytical and analytical issues. * Prepare NGS libraries (e.g., metagenomic or amplicon approaches) and perform target enrichment (hybrid capture) workflows. Quantify and QC libraries, sequence using Illumina and Oxford Nanopore approached, and perform first level analysis, QC, and interpretation of sequencing results. * Operate, monitor, and perform routine maintenance for sequencing instruments and supporting laboratory equipment. Document instrument performance and coordinate service as needed. * Plan and execute assay development and validation studies and summarize results in organized validation reports. * Maintain accurate documentation (lab notebook, SOPs, batch records, QC logs, incident reporting, and sample chain of custody) and follow institutional biosafety and data security requirements. * Support specimen biorepository operations including specimen receipt, processing/aliquoting, labeling, storage, inventory management, and sample and data tracking in laboratory information management systems. * Coordinate daily laboratory operations including reagent preparation, inventory management, ordering, and tracking of supplies. * Perform primary data review and interpretation for molecular and sequencing outputs; collaborate with bioinformatics partners and utilize platforms such as Terra and standard analytical tools for QC summaries and reporting. * Communicate effectively with supervisors and collaborators regarding assay status, issues, and timelines. These "first-of-a-kind" lung cancer models comprise the major discovery platform for projects throughout the lab and have enabled numerous therapeutic advances in the clinic. This position is ideal for someone who has recently completed their undergraduate studies (last 1-2 years) looking to expand their laboratory research skills in a translational research setting and while making contributions to the rapidly-evolving field of cancer therapeutics prior to attending graduate school. This opportunity is designed to provide an enhanced experience for prospective graduate (PhD) and medical (MD, MD/PhD) students during their gap years and provides excellent opportunities for leadership and career development, as well as co-authorship on manuscripts.
