What you will be doing:
The Predictive and Investigative Safety Assessment (PISA) group is seeking a highly motivated Intern to join our In Vitro Toxicology team located in Boston, MA. This individual will join a team of toxicologists working to design preclinical in vitro models to assess of the genotoxicity of novel therapeutic agents to de-risk potential drug candidates throughout the drug discovery process. The ideal candidate will have strong wet-lab skills and will be eager to learn or implement their computational skills to enable them to contribute to the experimental setup, implementation, and data analysis steps of the assay development process. Proficiency in various molecular and cellular technical skills is required. The applicant must be a demonstrated team player, adaptable to changing needs and able to work closely and responsively within their team. Excellent communication and people skills are required. The intern will learn about the drug development pipeline, assay development in industry, and the field of genotoxicity and related regulatory requirements.
* The successful candidate will:
* Contribute to the development of in vitro assays to support the selection and de-risking of potential drug candidates through close interactions with colleagues within PISA

The candidate will be located either at the cell and gene therapy CMO's site and/or be able to travel up to 50% of their time to those sites as a "Person-in-Plant" to oversee activities on the ground, and/or as a SME to provide technical support to resolve emerging issues. The successful candidate will be a key member of the Vertex/CMO Virtual Plant Team (VPT) contributing to the broader External Manufacturing group activities.
Key Responsibilities:
* Function as the first/primary point of contact for all Vertex interests/activities at ATMP cell and gene therapy Contract Manufacturing Organizations (CMOs)
* Provide operational oversight of, and troubleshooting support to, CMOs to ensure all deliverables meet or exceed Vertex and regulatory requirements (including but not limited to successful execution of manufacturing operations, on-time shipments, quality compliance and cost management); be Vertex's "eyes and ears" at the CMO for right-first-time execution of commercial operations.
* Proactively "connect the dots" on emerging trends and issues and lead/influence cross functional teams to drive resolutions and minimize impact to compliance and delivery of safe and reliable supply
* Maintain on-site presence at CMO facilities for extended periods (including off hours/weekends) to oversee (i) critical program activities, (ii) clinical and commercial batch manufacturing until the CMO is sustainably executing operations, and (iii) Health Authority inspections.
* Leverage Technical, Quality and Regulatory SMEs to provide guidance to CMOs and ensure cross-functional alignment on program priorities and deliverables from clinical through commercial manufacturing and supply.

* Identify and escalate business-critical issues to Commercial Manufacturing and Supply Chain, CMC, and Quality leadership.

* Lead continuous improvement initiatives to affect timely resolution of supply issues.

The Importance of the Role
The Director of EHS is a senior leader responsible for shaping and driving Sarepta's global EHS strategy. What you will be doing:
The In Vitro team within the DMPK department uses in vitro permeability assays to screen drug candidates and support the prediction of oral bioavailability via absorption, distribution, metabolism, and excretion (ADME) processes for in vivo-in vitro correlation (IVIVC). The intern position will work with the In Vitro team to method optimize and validate permeability assays to best predict oral bioavailability of small molecules and the expansion to beyond Lipinski's Rule of Five drug candidates.
Responsibilities include:
* In vitro PAMPA assay method optimization and validation with literature compounds at various assay conditions to achieve optimal assay performance, in relation to cell-based in vitro screening assays for best in vivo oral bioavailability prediction
* Exploration of novel in vitro based assays/techniques that mimic physiological state and in vivo formulation compatible to address questions, and for better prediction of bioavailability
* In vitro assay application and expansion to beyond Rule of Five molecules
* Data analysis, correlation, and interpretation compared to published sources
* Discuss and present research progress with In Vitro team, DMPK department, and stakeholders as appropriate
What you will need to succeed:
