Working under general supervision, the Research Assistant 2 will plan and carry out research on the effect of deleting the Glut1 glucose transporter on the progression of diabetic kidney disease. An objective of this position is to understand the molecular mechanisms and cell signaling pathways altered by inhibition of proximal tubule Glut1-dependent glucose uptake. This position will include work with animals (mice) and animal tissue.
ESSENTIAL FUNCTIONS
Plan and carry out small animal surgery independently, after instruction as required by experimental protocols. A small amount of animal husbandry, which could include administration of drugs, may be required. (25%) Conduct a series of experiments to supply scientific information as part of a larger project. Perform Western blots and RT-PCR analyses for Glut1 expression. This will require performing nephrectomy on anesthetized rats and collecting renal cortical tissue. May assist in developing improved techniques or procedures. (15%) Perform measurements of glomerular filtration rate (GFR). These experiments will require intravenous injection of fluorescently labeled compounds, and then fluorescence decay acquisition using a transdermal device. Collect and analyze data using proprietary software. (20%) Collect urine from mice and measure urine albumin by ELISA and urine creatinine by picrate methods.
