JOB SUMMARY:
The Lab Technologist is a motivated and detail-oriented individual with laboratory experience who also excels in communication, organization, and customer service. This role requires technical experience and interpersonal skills to contribute to efficient operation, effective teamwork, and excellent client interactions. The position involves the receiving, organizing, and logging of >50k samples annually in preparation for drying and grinding. The Lab Tech also assists in equipment maintenance, labware cleaning, and site organization. These critical sample handling and maintenance roles contribute to the overall laboratory goals of serving the agricultural community of SC. The Lab Tech assists with administrative tasks and other duties as assigned.
JOB DUTIES:
50% - Essential - Sample Receiving and Organization:
Organization and preparation of samples (feed, plant, water, animal waste, compost, soil, and research) by receiving shipments, sorting, logging in, drying, and grinding, following detailed procedures, and operating equipment, i.e., grinders, analytical balances, muffle furnace, and ovens, while following best practices for laboratory safety and sample integrity. Disposal of samples.
15% - Essential - Lab/Section Maintenance and Improvements:
Maintain general laboratory workspaces, glassware, and equipment for cleanliness and serviceability, including stocking supplies and forms for receiving area, maintaining organization
of area and samples. Use appropriate laboratory safety practices and procedures in all activities. Assists with annual maintenance/improvements of facility. Assists lab manager with maintenance on equipment as needed.

JOB SUMMARY:
The technician will be responsible for creating non-pathogenic DNA constructs to be used in single molecule biophysics experiments and other duties as assigned. Assembly of DNA fragments into plasmids using traditional restriction digest cloning, site-directed mutagenesis, Gibson assembly, or ligation independent cloning, etc. Non-pathogenic Escherichia coli bacteria will be transformed with the assembled plasmids to produce useful amounts to be used as PCR templates or for digestion and purification for single molecule experiments. Other duties as assigned.
JOB DUTIES:
60% - Essential - Use a variety of molecular cloning techniques to assemble DNA fragments into plasmids. Some fragments will be prepared from DNA sequences available in the lab and others will be purchased commercially. Successful assembly will be verified by gel electrophoresis and polymerase chain reactions.
30% - Essential - Once designed sequences of DNA have been assembled into plasmids, responsible for generating purified fragments of the plasmid sequences using restriction digests or polymerase chain reactions followed by purification and possibly ligation. Larger quantities of DNA may require salt/alcohol or salt/polyethylene glycol precipitation. The purified fragments will serve for experiments conducted by graduate students and postdoctoral researchers in the lab.
10% - Essential - Responsible for organizing storage of the DNA and protein aliquots available in the lab in -20 and -80 degree C freezer compartments and maintaining an updated database with location information.
MINIMUM REQUIREMENTS:
Education - A high school diploma and work experience in a clinical or research laboratory setting, in a health care setting or in a general maintenance or machine shop environment. A bachelor's degree in a related scientific discipline may be substituted for the required work experience.

Develop, draft, revise SOPs, training documents, validation/verification plans, test cases, and validation/verification summaries. This high complexity testing identifies chromosome abnormalities that assist in diagnosis, prognosis and/or guide treatment of genetic disease in constitutional disorders as well as somatic genetic alterations in cancer, primarily hematologic disease.
Independently perform chromosome analysis for genetic disease (hematologic and constitutional)
Perform cell culture work - set-up, harvest, slide preparation, slide banding, FISH setup and reagent preparation for a variety of specimen types
Assure integrity of specimens throughout all phases of workflow
Perform work with the highest standard for accuracy and precision.
Understand the basis of each test. Use knowledge to make critical judgements regarding acceptability of test results.
Troubleshoot instrumentation and culture/harvest issues
Maintain and adhere to accurate documentation for QC/QA logs

Summary:

25% - Prepare and maintain records for all procedures performed to facilitate reporting to investigators and the NIH annual report. Respond to requests for information and queries pertaining to processing procedures and records in a courteous and timely manner.
15% - Adhere to laboratory safety standards and assist in maintaining overall laboratory organization and supply inventory. Assist in the development and implementation of policies, procedures and protocol written instructions to ensure compliance with regulatory standards.
10% - Perform complex mathematical calculations required for laboratory methodology, including reagent preparation and dilutions. Perform laboratory procedures, including procedures requiring aseptic technique or special handling methods. Assists in performing qualitative/ quantitative tests.
