Location/Division Specific Information
You will be a part of the Applications Development team within the Life Sciences Business Unit (LSBU) of Thermo Fisher Scientific's Material and Structure Analysis Division (MSD). The LSBU provides high-value Electron Microscopy workflow solutions for high resolution biological imaging, ranging from atomic resolution imaging of protein complexes to volume acquisition of cellular compartment structures.
The position is embedded in the Center of Excellence for Structural Cell Biology at St. Jude Children's Research Hospital in Memphis, Tennessee. The Scientist will operate from this location.

How will you make an impact?
You will be responsible for development of pioneering methods and protocols for cryo FIB milling with plasma FIB (pFIB) within the electron Cryo-Electron Tomography (Cryo-ET) and Volume EM workflows, using our groundbreaking Arctis and Hydra Bio cryo-plasma-FIBs. You will coordinate applications development for our cryo-FIB projects with our collaborators in a world-class cryo-imaging lab. Your role will involve cryo FIB milling to prepare cellular lamella and optimizing methods for sample preparation, with the ultimate goal of performing cryo-Electron Tomography on our Krios cryo-Transmission Electron Microscope (TEM) instruments. Additionally, you will develop methods and workflows for volume imaging of high pressure frozen cellular and tissue samples using cryo Auto Slice & View and related technologies on the pFIB dual beam systems. You will build a network within the scientific community through collaborations with new and leading customers and help shape the future of cryo EM.
Deliverables and responsibilities:
Develop and evaluate FIB milling and volume imaging strategies with plasma FIB on cells and tissues. Work with collaborators for training users (on site and remotely) and develop and implement proof of concept experiments with challenging samples. Your contributions will support directly the customer's scientific endeavor as well as feed directly into new product development at Thermo Fisher.

Support R&D, product development, marketing, and sales by lending applications expertise.
Communicate new applications results at scientific workshops, conferences and contribute to scientific publications.
Serve as an expert for cryo EM sample preparation with plasma FIB instruments
Provide feedback to engineering and software development teams on system performance, new design features, new hardware qualification and advanced use-case problems for milling and cryo ET data collection.
Provide marketing and sales materials to the Product Marketing team for integration into training programs.
Make significant contribution to defining the framework and future direction of cryo TEM products.
How to get here:
This position is ideal for an expert in focused ion beam (FIB) electron microscopy, ideally with a familiarity in both cryo-FIB milling and TEM sample preparation. Strong technical understanding of FIB theory and technology as well as experience with Plasma FIB instrumentation is a crucial requirement. A background or experience with biological questions and samples is desirable but not a requirement for the role.
Education, experience, skills, abilities:
An advanced degree (e.g .PhD, MSc) in the fields of Physics, Biophysics and related or equivalent fields.
Demonstrated in-depth experience in TEM sample preparation with plasma FIB milling as well as a deep understanding of FIB technology. Knowledge of soft matter samples, with cryogenic conditions and biological specimen is a plus.
Having prior experience with TEM instruments is advantageous.
Experience in interacting with new users as well as highly technical customers and the ability to communicate complex technical concepts to a variety of audiences.
Demonstrated ability to systematically develop new and improve methods, protocols, troubleshooting techniques and operational optimization.
Excellent verbal and written communication skills with a diverse audience.
Ability to independently identify, define and implement solutions and to own and lead projects.

The lab of Jeremy Chase Crawford is seeking a highly skilled bioinformatician to lead the computational analysis of next-generation immune repertoire sequencing data. This position will work at the cross-section of immunology and computational biology to decipher T-cell specificity in the context of vaccine responses (e.g., SARS-CoV-2) and pediatric immune development.
The successful candidate will be responsible for developing robust automated pipelines for novel assays-specifically TIRTLseq and 5'RACE-to track clonal expansion, infer HLA genotypes, and map TCRs to their cognate epitopes.
Key Responsibilities
Pipeline Development & Automation:
Develop and maintain robust pipelines to automate the parsing, quality control, and characterization of human sequencing data.
Create automated reporting tools to characterize clonal expansion across longitudinal samples and assess sensitivity differences between assay platforms.
Advanced TCR Repertoire Analysis:
Perform paired chain (alpha/beta) analysis to infer patient HLA genotypes and "de-orphan" single-chain TCRs from earlier time points.
Utilize network analysis approaches to infer the epitopes recognized by expanding TCR clones.
Database Management:
Create and maintain databases to track subject and sample metadata, ensuring data integrity for any possible audits.
Merge wetlab and computational data sources (LIMS, Spreadsheets, devices, bioinformatic pipelines etc.) to create up-to-date reports in PoweBI.
Design and optimize a static and iteratively updated TCR/epitope matching database to predict antigen specificity.
Evaluate data for quality assurance and mine relevant bioinformatics databases to support wet-lab validation.
Collaboration & Communication:
Work closely with wet-lab scientists to design experiments (e.g., defining sample exclusion criteria based on expansion evidence) and interpret complex immunological data.
Assist in the evaluation of products and solutions pertaining to St. Jude's technology needs (e.g., cloud computing integration).
Minimum Qualifications
For Senior Bioinformatics Analyst:
Bachelor's degree with 2+ years of relevant post-degree work experience (or Master's degree with 0-2 years experience) in Bioinformatics, Computational Biology, or a related field.
Experience in at least one programming language (Python preferred) and statistical package (R).
For Bioinformatics Research Scientist:
Bachelor's degree with 4+ years experience, Master's with 2+ years, or PhD with no work experience.
Demonstrated ability to independently perform advanced statistical analysis and algorithm implementation.
Preferred Skills for this Project
TCR Specificity: Experience with immune repertoire tools (e.g., MiXCR, VDJtools, TCRdist) and handling paired-chain sequencing data.
Structural Biology/Inference: Familiarity with HLA inference algorithms and epitope prediction models.

About the Role:
Chief responsibilities include gynecologic oncologic surgical care of patients along with outpatient and inpatient consultation. This position is also responsible for training medical students, Residents, and Fellows. This position includes the opportunity for protected research time, and successful applicants are expected to both participate in and serve as site Primary Investigators on clinical in the Department of gynecologic oncology. There is also the opportunity for collaboration with other investigators in an exciting clinical research environment. This Physician will provide care in a clinic setting, offering state-of-the-art surgical options to patients (access to over 12 robotic Xi) to treat both malignant and benign diagnoses. The ideal candidate will be board certified/board eligible and possess excellent clinical and surgical training. We focus on high-volume clinical care, accessing the best education, research, and clinical care. We are laser-focused to bring the best care available to our communities of practice while focusing first on the balance of life that is mandatory for our gynecologic team. You will have the opportunity to work with a talented interdisciplinary team of scientists to develop a cellular therapy with the goal of treating patients here at St. Jude Children's Research Hospital and worldwide. The ideal candidate will have a strong combination of experience with genome editing and human hematopoietic stem cells (HSCs).
You will also have the opportunity to learn about and support new, exciting projects, including the development and optimization of pre-clinical genome editing strategies to treat hemoglobinopathies and other hematological diseases, process development, and IND-enabling studies (Pharm/Tox).
Job Responsibilities:
Perform studies to develop genome editing strategies for human hematopoietic stem cells (HSC).
Design and execute IND-enabling experiments to characterize the pharmacology of a genome-edited human hematopoietic stem cell product to treat sickle cell disease.
Characterize the efficacy, safety, and potency of the HSC cellular drug product in
in vitro
and xenotransplantation assays.
Multicolor flow cytometry and FACS to sort hematopoietic populations of relevance.
Develop experimental methods including RT-PCR, dd PCR, next-generation sequencing, and single-cell sequencing.
Perform NGS library preparation and analysis of editing outcomes, on-target and off-target.
Evaluate scale-up and close-system processes to ensure reproducibility of the drug product.

What problem are we tackling?
Target discovery is a critical foundation of drug development, underpinning efforts to overcome the persistent challenges of drug resistance, emerging pathogens, and complex diseases that lack effective therapies. By uncovering novel proteins and molecular pathways driving disease progression, we can enable the rational design of more selective interventions that reduce off-target effects and improve patient outcomes. The development of tool molecules, coupled with detailed characterisation of their inhibitory mechanisms, provides a direct bridge between phenotypic observations and small-molecule drug design, accelerating the translation of basic discoveries into clinically actionable therapies.
What is our approach?
Short Linear Motifs (SLiMs) are compact functional modules embedded within the intrinsically disordered regions of proteins. They mediate a wide range of regulatory processes, including the assembly of dynamic complexes, recruitment of modifying enzymes, control of protein stability, and specification of subcellular localisation. Despite their central role in cellular regulation, SLiM-binding pockets remain a largely untapped class of therapeutic targets. By applying large scale base-editing and peptide inhibitor studies the project will address three key questions: (i) How essential are SLiM-mediated interactions for cellular fitness? (ii) Does SLiM essentiality differ between healthy and diseased cells? (iii) Can these interactions be selectively targeted for therapeutic intervention?
