Research in the Zhang Laboratory focuses on exploring the function of double-negative T (DN T) cells in antitumor immunity and cancer immunotherapy. DN T cells play a pivotal role in initiating antitumor immune responses and shaping the tumor microenvironment. Understanding the underlying mechanisms will enable the development of a novel paradigm for cancer immunotherapy. The successful candidate will play an integral role in advancing cutting-edge research funded by an NIH/NCI R01 grant.

**How will you make an impact?**
You will be responsible for development of pioneering methods and protocols for cryo FIB milling with plasma FIB (pFIB) within the electron Cryo-Electron Tomography (Cryo-ET) and Volume EM workflows, using our groundbreaking Arctis and Hydra Bio cryo-plasma-FIBs. You will coordinate applications development for our cryo-FIB projects with our collaborators in a world-class cryo-imaging lab. Your role will involve cryo FIB milling to prepare cellular lamella and optimizing methods for sample preparation, with the ultimate goal of performing cryo-Electron Tomography on our Krios cryo-Transmission Electron Microscope (TEM) instruments. Additionally, you will develop methods and workflows for volume imaging of high pressure frozen cellular and tissue samples using cryo Auto Slice & View and related technologies on the pFIB dual beam systems. You will build a network within the scientific community through collaborations with new and leading customers and help shape the future of cryo EM.
**Deliverables and responsibilities:**
Develop and evaluate FIB milling and volume imaging strategies with plasma FIB on cells and tissues. Work with collaborators for training users (on site and remotely) and develop and implement proof of concept experiments with challenging samples. Your contributions will support directly the customer's scientific endeavor as well as feed directly into new product development at Thermo Fisher.

**How to get here:**
This position is ideal for an expert in focused ion beam (FIB) electron microscopy, ideally with a familiarity in both cryo-FIB milling and TEM sample preparation. Strong technical understanding of FIB theory and technology as well as experience with Plasma FIB instrumentation is a crucial requirement. A background or experience with biological questions and samples is desirable but not a requirement for the role.

+ An advanced degree (e.g .PhD, MSc) in the fields of Physics, Biophysics and related or equivalent fields.

LIDL's work is deeply collaborative, involving partnerships with federal sponsors; disciplines like coastal engineering, hydrology, ecology, and geomorphology; and local communities, as well as other landscape architects. Currently, we are working on a broad range of coastal infrastructure projects from the Great Lakes south to the Gulf of Mexico, sponsored by funders including the US Army Corps of Engineers' Engineering With Nature program, the National Oceanic and Atmospheric Administration's Effects of Sea Level Rise program, and the Gulf Research Program of the National Academies of Sciences, Engineering, and Medicine. The Scientist-QC is part of the Lilly Lebanon API Quality Control team and provides technical leadership and expertise in the development, implementation, and maintenance of Quality Control laboratory systems to the Lilly Lebanon API site throughout the start-up, implementation, and day to day operations.
The Scientist-QC performs accurate and timely testing of routine and non-routine lab samples in accordance with appropriate GMP and safety guidelines. The Scientist completes second person verification of other's results. DEPARTMENT OVERVIEW
Dr. Richard M. Myers is a Faculty Investigator and Chief Scientific Officer of the HudsonAlpha Institute for Biotechnology. His laboratory uses genomic, genetic, molecular biology, and cell biology approaches to study problems in human neurobiology and gene regulation on a genome-wide level. We especially seek to understand how germline and somatic DNA sequence variation, epigenetics, and the environment affect the levels of transcription of genes in cell-type specific manners in the human nervous system. We focus on normal, healthy brain function as well as diseases, including Huntington disease, Alzheimer disease, Parkinson disease, psychiatric disorders, and amyotrophic lateral sclerosis, often in the context of differential responses to therapeutic agents. In addition, we work on neuropsychiatric disorders, and with Dr. Greg Cooper's lab at HudsonAlpha, on understanding the causes and mechanisms of unexplained diseases in children. Our experimental and computational work include genome-scale analysis of cis-acting regulatory sequences, DNA binding proteins and epigenetic action involved in human gene regulation and disease, as well as the development of molecular biomarkers for early detection and monitoring of diseases relevant to neuroscience.
While not required, subject area familiarity and/or interest in genetics, genomics, neuroscience, neuropsychiatric diseases, and/or neurodegenerative diseases is desirable. Project areas include study of gene regulation for genes implicated in normal human biology as well as disease, novel analysis approaches for associating genetic variation with gene expression, and genome-wide approaches relevant to disease as well as genome function in the brain more broadly.
POSITION SUMMARY
This Research Associate (RA) position is in the Myers Lab supporting genomics research projects in a variety of basic and applied areas, including global genomic assays and functional characterization. The activities of the RA include: molecular techniques including restriction digests, ligations, annealing, gel electrophoresis, quantification of nucleic acids, bacterial cloning, nucleic acid extractions; mammalian cell culture using fully consented cell lines; preparation of samples for next-generation sequencing; and other cellular and molecular biology protocols. This position requires exacting attention to detail, strong organizational skills, and the ability to work as part of a team as well as independently.
The Research Associate carries out research protocols as determined by the Principal Investigator. The Associate follows procedures exactly as outlined, ensuring the consistency of research practices and, ultimately, helping ensure the consistency and integrity of project outcomes. The Associate also provides feedback in the case that procedures are not producing expected results.
The Research Associate must understand laboratory procedure and safety. In addition to a general knowledge of the overall objectives of the research project, the Associate must have an understanding of the tactical methodology in particular. The Associate works under the direction of a graduate student, postdoctoral fellow, research assistant with supervisory responsibilities, senior scientist, or the lab Principal Investigator, communicating progress and advising of any issues that arise. This is the reason why the Valeo Power division is willing to implement "Design 4R" guidelines (4R standing for Robust Design, Repair, Remanufacture, Recycle) in order to make the next generation of products sustainability driven (Reman 2.0).
In the interest of efficiency, the development of a "4R Configurator AI Agent", acting as a Sustainable Design Guidelines Provider, will save development efforts in the OEM and Reman products development.
