By 1912, the school had moved to an opera house in Los Angeles and had grown to 113 students.
In 1922, the Chiropractic Initiative Act of 1922 passed, allowing the state to create a board of chiropractic examiners.
By 1950, the course of study had expanded to four years and the College moved to Glendale, California, consolidating its basic science subjects and chiropractic sciences into one comprehensive curriculum taught in one modern facility.

In April 2017, CUSM expanded its WASC Senior College and University Commission (“WSCUC”) eligibility to include the MBS program.
The first cohorts of 64 Medical Doctor (M.D.) program students started in July 2018.
CUSM welcomed 36 MBS program students in the summer of 2018.
In March 2020, the COVID-19 pandemic affected CUSM to switch gears and begin a virtual approach to learning.
First and second-year students have resumed limited in-person instruction for critical hands-on training in safe, secure small group settings during the fall of 2020.
As of 2020, the university continued to grow, adding two new masters programs; Master of Science in Medical Science and a Master of Acupuncture and Chinese Medicine.
