    The Robert M. Berne Cardiovascular Research Center was established to expand basic, translational, and clinical research of cardiovascular diseases. Cardiovascular diseases, including congestive heart failure, hypertension, atherosclerosis, stroke, aneurysms, and vascular auto-immune diseases are the most common cause of death and disability in the United States, and account for approximately 30% of deaths and health care costs annually in this country and developed societies. The mission of the Robert M. Berne Cardiovascular Research Center is to support interdisciplinary research in basic, translational and clinical cardiovascular sciences, including cardiovascular development, function, pathophysiology, pharmacology, genetics, genomics, and proteomics, and to apply this knowledge to better understand the causes of cardiovascular diseases and to pioneer development of new innovative therapies and approaches to prevent or treat them Position Summary The Robert M. Berne Cardiovascular Research Center (CVRC) is currently seeking a Research Associate to to conduct research on identification and molecular investigation of heart-derived secreted proteins within the Khetarpal Laboratory. The Khetarpal Laboratory multidisciplinary approach that combines mouse models (exercise, heart failure, dyslipidemia), viral vector-mediated gene delivery (AAV/adenovirus), primary cell culture, quantitative proteomics and lipidomics, CRISPR genome editing, and single-nucleus RNA-seq. Trainees will also have opportunities to connect findings to large-scale human genetic and proteomic datasets. The incumbent will participate in a multiplicity of relevant lab projects and in writing study reports for the study sponsors, papers, and grant applications and is expected to develop research programs as time permits. The incumbent will also be expected to attend and participate in laboratory meetings and CVRC-related seminars, as well as work with graduate students, technicians, fellows and collaborators in the laboratories involved in the project. Note: Research Associate positions are for candidates who have within the last 3 years. If you have more than 3 years of postdoc experience, in the field of study, you will not be considered. Responsibilities * Full-time research or scholarship at the University. * Preparatory for a full-time academic or research career. * Supervised by a senior scholar; and allows the appointee to publish the results of his/her research or scholarship. * In addition to the above job responsibilities, other duties may be assigned. Minimum Qualifications * Education: Doctoral degree Preferred Qualifications Experience in mouse metabolic and cardiovascular physiology (including exercise testing, echocardiography), molecular and cellular biology, and mammalian cell culture, are highly preferred. Particularly valuable additional experience includes: * Mitochondrial functional assays in cells and tissues (Seahorse, Ouroboros) * Culture of rodent cardiomyocytes, adipocytes, and endothelial cells * Familiarity with lipoprotein metabolism and lipoprotein metabolic studies in mouse models * Genome editing (CRISPR/Cas9) and viral vector transduction (adenovirus, AAV) * Analysis of proteomic, transcriptomic (bulk or single-cell), and lipidomic data * Experience with culture of induced pluripotent stem cell-derived cardiomyocytes (iPSC-CMs) Physical Demands This is primarily a sedentary job involving extensive use of desktop computers. The ideal candidate will have previous basic science laboratory experience including rodent handling and colony management, rodent survival surgeries, auditory function testing, neurophysiology, microscopy, complex statistical analysis and coding in MATLAB/Python. The lab's research focuses on understanding neural circuits in the descending auditory system, specifically how auditory brainstem neurons encode acoustic information and identifying/characterizing neuromodulatory (e.g. serotonergic) pathways that influence sound processing in the brain and cochlea. Using normal hearing and noise-induced hearing loss models, our long-term goals are to (1) improve basic knowledge of brain processes that modulate sound encoding by the cochlea and (2) identify translatable therapeutic otoprotective targets to reduce susceptibility to hearing disorders resulting from acoustic overexposures (i.e. noise trauma). This position offers a unique opportunity to be part of a collaborative research network spanning both The University of Texas at Dallas (UTD) and The University of Texas Southwestern Medical Center (UTSW). Located in the brand-new Texas Instruments Biomedical Engineering and Sciences (TI-BMES) building, the successful applicant will have access to cutting edge equipment/technologies through core facilities at both UTD Richardson and UTSW Dallas campuses in addition to those contained within the lab. As part of the Department of Speech, Language and Hearing, the lab works closely with other basic science laboratories and is uniquely positioned to collaborate with clinicians and audiologists to bridge basic science and clinical research. The post-doctoral fellow will be responsible for leading research using rodent (mouse) models, consisting of neuroanatomical circuit mapping (survival surgeries, brain/cochlear tissue preparation, immunohistochemistry, microscopy), in-vivo sound evoked neurophysiology (auditory function testing, single unit electrophysiology), in-vitro whole cell patch-clamp electrophysiology from brain slices (pharmacology), optogenetics and animal behavior. Our surgeons perform almost 20,000 operations per year for pediatric and adult patients. Training the next generation of surgeons and clinician-scientists is integral in our department, with more than 100 residents and fellows trained each year. The projects are focused on determining how tumor associated macrophages and microglia acquire alternatively activated states in pHGG and identifying novel therapeutic vulnerabilities to reinvigorate the anti-tumor immune response. Routine methods used in the lab are flow cytometry, immunohistochemistry, next generation sequencing, and primary cell cultures among others. Specifically, we will be focusing on: 1.) Defining molecular changes that occur within reactive astrocytes in Down Syndrome and Alzheimer's Disease, 2.) Targeting astrocytes to improve neuronal synaptic dysfunction, 3.) Understanding reactive astrocyte-microglia and astrocyte-vascular crosstalk in disorders of the nervous system. Required skills include animal handling, experience in confocal imaging and immunohistochemistry, and experience in wet lab techniques such as western blotting, RNA extraction, and molecular cloning. Desired skills include experience with proteomics and familiarity with bioinformatic analysis pipelines, as well as exposure to iPSC differentiation into neuronal and glial cells. Postdoctoral employment is temporary and is normally limited to an individual who has been awarded a Ph.D. or equivalent doctorate within the previous five years1 and who will be involved in full-time research or scholarship at the University.
