Research Technician, Molecular Physiology and Biophysics
Vanderbilt University 4.6
Researcher job at Vanderbilt University Medical Center
The research technician will work in Dr. Nancy Carrasco's laboratory in the Department of Molecular Physiology and Biophysics (MPB) at Vanderbilt University School of Medicine. The research technician will be responsible for experiments conducted in the laboratory, including: mouse husbandry, tissue culture, recombinant DNA studies, site-directed mutagenesis, microscopy (both light and electron), electrophysiology, transport assays, protein purification, western blots, and cryoelectron microscopy (CryoEM). This position will require scientific excellence, well-honed experimental skills, accuracy, the ability to conduct research according to a predetermined schedule, and the ability to report experimental outcomes to the supervisor (i.e., the lab head) effectively. In addition, this position requires skill at record-keeping and basic computer skills such as the ability to use Microsoft Word, Excel, and PowerPoint. The successful candidate must commit to remaining in the position for a minimum of two years. Therefore, the position is suitable for an entry-level scientist aiming to gain substantial experience in experimental research in preparation for applying to graduate programs. The laboratory has an excellent track record of producing scientists who have been admitted to PhD, medical, and MSTP programs at top universities.
The Carrasco laboratory focuses on pushing forward our understanding of the action of thyroid hormones-from their biosynthesis in the thyroid gland to their pleiotropic effects on many different tissues. We express and purify wild-type sodium/iodide symporter (NIS) protein and mutant NIS proteins engineered by site-directed mutagenesis; ascertain the effects of the relevant mutations on NIS function; and determine the atomic structures of the mutant proteins using cryoelectron microscopy (cryo-EM). We also use NIS knockout mice to investigate the metabolic effects of varying degrees of hypothyroidism, and we are currently investigating novel approaches to employing not only wild-type NIS but also NIS mutant proteins in gene-transfer studies to image and treat several cancers. We value excellence, commitment, motivation, hard work, creativity, curiosity, and passion for science. Our overall experimental approach runs the gamut from engineering NIS cDNA constructs, expressing proteins in mammalian cells, membrane protein purification biochemistry, high-resolution structure determination using cryo-EM, image processing, light microscopy, and electrophysiology to investigating the molecular mechanisms of wild-type and mutant NIS proteins. The results of our research deepen our understanding of sodium-driven active transport and molecular endocrinology more generally, which may ultimately lead to translational approaches to treating thyroid disease and several cancers.
Duties and Responsibilities
Conducting research to their, and our, highest standards, as well as to the highest possible standards of responsible conduct of research.

Research Assistant, Biochemistry, Karbstein
Vanderbilt University 4.6
Researcher job at Vanderbilt University Medical Center
About the Work Unit: The Karbstein Lab in the Department of Biochemistry at Vanderbilt University is seeking an enthusiastic molecular biology/biochemistry research assistant to aid in the lab's studies of ribosome assembly, quality control and repair. In addition to providing a better understanding about the cellular mechanisms underlying ribosome integrity, this basic science work is highly relevant to cancer biology, the biology of aging. The lab relies heavily on biochemistry and molecular biology, as well as yeast genetics. For examples of the type of research that will be undertaken see: Blomqvist et al, 2023, PLoS Genetics; Yang et al, 2023, Mol Cell; and Parker et al, 2024, PLoS Biology; Yang&Karbstein, BioRxiv 2024.
The lab is a mid-sized lab seeking a research associate who will spend their time performing research, with some time spent maintaining lab infrastructure. The new research associate will be directly mentored and instructed by Dr. Karbstein, the Principle Investigator, and plan, conduct, interpret and present research. The laboratory is a supportive, fast-paced environment for driven candidates who enjoy a collaborative atmosphere. It is expected that the new research associate will produce enough data for a first author publication within 1-2 years of joining the lab.
The lab strives to be a diverse, inclusive, and equitable working environment; prioritizes personal growth and wellness; and is committed to work-life balance. Overall, this is an excellent opportunity for an experienced researcher who enjoys the independence of an academic lab.
Key Functions and Expected Performances:
Research and Development:
* Purifies ribosomes from various organisms for mass spectrometry analysis
* Experience in RNA biochemistry is an asset
* Uses protein gels, Western blotting and mass spectrometry to analyze ribosome assembly factors and ribosomal proteins
* Uses Northern blotting to analyze ribosome assembly in cells
* Uses agarose gels to analyze PCR screens and PCR reactions
* Generates yeast and worm strains and human cell lines
* Uses quantitative growth measurements to monitor genetic interactions
* Designs and clones proteins and RNAs for yeast, worm or human in vivo work and in vitro assays
* Analyzes RNAseq data
* Analyzes experiments independently and prepares them for presentation to the PI
Supervisory Relationships:
* Might supervise interns, work-study students or undergraduate researchers.
* The position reports administratively and functionally to the PI
Education and Certifications:
* A Bachelor's degree in cellular biology, molecular biology, biochemistry, or a related field from an accredited institution of higher education and a minimum of four years of experience is required.
Experience and Skills:
* Ability to perform all basic calculations related to buffer composition (Molarity, dilution factor, etc.) is necessary.
* Experience with protein and/or RNA biochemistry is strongly preferred.
* Computational experience is preferred.
* Being comfortable in learning new assays is necessary.
