This research opportunity is with the McElrath Lab within the Vaccine & Infectious Disease Division. The technician will work under the direction of a Staff Scientist managing the molecular profiling team in support of the antigen-specific B cell platform to help with the characterization of antigen-specific B cell receptor repertoires in the context of HIV vaccine trials. In particular, they will assist in developing and implementing high-throughput B cell receptor sequencing assays, including BCR PCR, Illumina NGS library preparation, CITE-seq, micro neutralization screening, PCR cloning and transient transfection for monoclonal antibody production and functional screening of these monoclonals. They will also be responsible for performing a variety of molecular and cellular biology techniques including multiplex PCR, single-cell RT-PCR, digital droplet PCR, maintenance of feeder cells under aseptic conditions, and RNA/DNA isolation. They will be involved in data management, interpretation and reporting on experimental results and assist with data presentation for lab meetings, conferences and manuscripts.
This person should be capable of maintaining detailed laboratory records, analyzing data, and reporting experimental results. They will interact closely with other laboratory staff to coordinate both up and downstream processing of samples and they need to a be a good team player. They will assist in the development and maintenance of laboratory quality control (QC) methods and records, which may require adherence to GCLP (Good Clinical Lab Practice) standards.
Responsibilities
* Perform molecular biology techniques: isolation of RNA/DNA from human lymphocytes for downstream analysis, gene expression by high density multiplex PCR, RT-qPCR, RNA-Seq, RT-PCR amplification for targeted amplicon sequencing of B cell receptor genes (paired and unpaired), including indexing and library preparation for NGS Illumina platform, antibody cloning and purification, monoclonal antibody production by transient transfection.
* Perform tissue culture techniques: maintenance of cell lines including frozen inventories, transduction of feeder cells with retroviral and lentiviral vectors, virus neutralizations, cytotoxicity assays, in vitro cell manipulations, including peptide stimulations and co-cultures.
* RNA and DNA isolation from whole blood, PBMC and tissue, Library preparation and indexing for Illumina sequencing platform
* Operate and carry out basic maintenance and QC of laboratory equipment.
* Perform and/or optimize new assays and specimen processing methods as needed.
* May prepare stocks of HIV pseudoviruses for downstream neutralization.
* Prepare and maintain reagents, stocks, and cultures necessary to perform the work.
* Independently collect and analyze data.

Qualifications
MINIMUM QUALIFICATIONS:

Vision Bioenergy Oilseeds (VBO) is a agricultural joint venture with the mission of developing, growing and marketing high quality oilseed oil for use in the production of biofuels to help create a cleaner, more sustainable future. We operate a research & development center and processing facility in Nampa, Idaho.
The Research Technician will ensure the Research Station, functions efficiently by performing a variety of duties to maintain and operate the facilities, equipment and farmland. Supports the Station Manager and other scientists on station with scheduling and resourcing research activities and ensuring efficient, functional execution of research activities. Support off-station field plot location growing activities. Primary crops under development include camelina and other oilseeds crops for the renewables energy markets and dietary supplement for livestock feeding markets.
The successful candidate will have:
Bachelor's degree in Agriculture preferred, or up to 5 years of related ag production or field research experience.
Ability to obtain and maintain an Idaho Private Applicator License (within 3 months of hire).
Experience with agricultural practices including soil and irrigation management, fertility, weed control, and safe equipment operation.
Proficiency operating field equipment (e.g., tractors, planters, sprayers, harvesters) and ability to work with GPS/electronic controls.
Basic equipment and facility maintenance skills.
Proficiency with Microsoft Office (Excel, Word, PowerPoint) and willingness to learn internal systems.
Valid driver's license with a satisfactory driving record.
Ability to work extended hours during the growing season (April-September), including rotating weekends; occasional evenings, weekends, overnight work, and travel.

MORE DETAILED RESPONSIBILITIES:
Conduct experiments, to completion, according to specific procedures, exercising independent judgment and discretion as appropriate.
Maintain healthy plants, collect accurate and timely data.
Maintain, organize and summarize results of experiments, noting problem areas, while recommending actions to be taken within standard procedures.
Organize and supervise work of temporary employees and station regular employees ensuring work is performed in safe and efficient manner.
Interact with state, local and federal authorities to ensure compliance with relevant regulations.
Manage field plot rotation, field plot health, and timely management for all plots.
Assist in conducting experiments at other research sites, laboratory areas, or groups.
Maintain appropriate inventory of chemicals and/or supplies necessary for the completion of experiments at work location.
Troubleshoot plant or mechanical issues that arise.
Operation and care of field equipment.
Operate and maintain irrigation equipment.
Perform other duties as assigned by management.
Understand and follow all QMS/RMS/HSSE protocols, policies and procedures.
Ensure that work is performed in a safe and efficient manner.
Report any accidents, incidents, near misses and hazardous observations to your Supervisor immediately.
Be proactive about facility care and cleanliness.

EDUCATION/KNOWLEDGE/EXPERIENCE:
Bachelor's degree in agriculture is preferred, but not required, and/or up to 5 years' industry related experience in ag production or ag field research activities.
Required to hold and maintain a private applicators license from the state of Idaho. Must obtain within three (3) months of hire.
In-depth understanding of agricultural cultural practices (e.g., ground preparation, soil and irrigation management, weed control, fertility management), and safe equipment operation.
A good understanding of Microsoft programs such as Excel, Word and PowerPoint and ability to learn internal programs.
Knowledge and experience of maintenance of equipment and facilities.
Previous experience operating various field equipment, including but not limited to tractors, harvesters, tillage, planters, spray equipment, soil and fertilizer applications. Computer proficiency for GPS tractor operation and other equipment requiring interaction with electronics.
Knowledge of agricultural practices, safety procedures and greenhouse operations.
Work schedule may require longer hours based on Ag season (April-September). Rotating weekends (partial day Saturday/Sunday).
Some evenings, weekends, and overnight work (and travel) may be required.

The Sahoo Lab at the Fralin Biomedical Research Institute studies the genetic and functional mechanisms that drive bone marrow failure, clonal hematopoiesis, and progression to MDS and AML. We integrate genomic and functional approaches on patient derived samples, iPSC and mouse disease models to uncover mechanisms and therapeutic vulnerabilities.
Current project areas include:
1. Germline predisposition to MDS and AML: mechanisms by which SAMD9L mutations cause hematopoietic failure and predispose to leukemogenesis
2. Chromosome 7 aneuploidy syndromes: modeling monosomy 7 associated bone marrow failure and its role in clonal selection
3. Clonal evolution and therapeutic vulnerabilities: using single cell sequencing to track clonal dynamics and to identify targets in high risk MDS and AML
We seek a full-time Research Tech to support our cellular, and in vivo studies. The role requires strong organizational skills, careful record keeping, and comfort working in a fast-paced research environment.
The key responsibilities will include if not limited to:
• Perform molecular biology workflows: DNA/RNA extraction, cloning, standard PCR genotyping, qPCR, gel electrophoresis.
• Support mouse studies: Main mouse colonies, perform genotyping, manage timed breeding, performing procedures (tail vein and intraperitoneal injections, routine blood collection) and sample collection (per approved protocols).
• Participate in laboratory projects involving the application of basic molecular biology techniques and mouse animal models.

Duties may include:
* Provide technical support in planning and/or designing of scientific projects, studies, or applications
* Plan and conduct research and analysis of scientific data
* Interpret test results and comparing with determined standards and limits
* Identify and evaluating scientific samples and determining need for further testing
* Develop recommendations and/or providing solutions, standards, and protocol to a wide range of difficult problems
* Plan, design, develop, monitor and/or evaluate portions of a scientific project, study, or application
* Provide scientific evaluations/assessments
* Research, implement, standardize, and evaluate new procedures
* Maintain detailed records of results and prepares written reports acceptable to the Principal Investigator and funding agencies
* Other duties as assigned
