This position works closely with the Principal Investigator to identify the endogenous molecular factors and downstream signaling mechanisms regulating arterial lymphatic vessel formation and their role in the development of atherosclerosis, hepatic steatosis, and other cardiometabolic diseases. The project involves the use of multiple genetically engineered mouse models, in-vivo gene transfer, molecular biology techniques, as well as various biochemical, functional, and cell biology assays. MINIMUM REQUIREMENTS: Doctoral Degree. KNOWLEDGE, SKILLS, AND ABILITIES: Strong written and oral communication skills Highly motivated and productive. Ability to perform experiments independently after minimal training. The Abraham lab studies gene expression-regulation mechanisms. We are recruiting computational biologists to collaboratively develop software approaches to analyze high-throughput sequencing (-omic) data. We build analytical software pipelines to find answers to biological questions about gene regulation in genome-wide datasets, usually from applied sequencing experiments like CUT&RUN, RNA-Seq, and HiChIP, as well as single-cell omic experiments. Our interests center on enhancers and super-enhancers. Specifically, we seek to understand how these regulatory elements establish gene expression programs in healthy cells, and how enhancers are altered by mutation, abused by mistargeting, and targetable with drugs in diseased cells. We characterize the specific core regulatory circuitries driving disease-relevant cells and seek to understand how mutations in the non-coding DNA of such cells can drive disease, including cancers, through gene misregulation. The successful candidate will become a fundamental component of a multidisciplinary, inter-institutional team assembled to study how gene expression regulation meaningfully differs between normal and pediatric cancer cells and might be productively targetable. The successful candidate will lead research projects within the laboratory with increasing independence in daily operation. Ideal candidates will have experience building, tailoring, and deploying analysis pipelines using widely available genomic analysis toolkits (e.g. bedtools, samtools, HiCPro, Tuxedo tools), as well as experience managing large numbers of datasets. Our laboratory conducts behavioral and 2-photon laser-scanning microscopy imaging of neuronal activity in mutant mice, with a focus on models of high-risk genetic risks for schizophrenia. Our laboratory conducts 2-photon laser-scanning microscopy imaging of neuronal activity in the auditory system of behaving mice and rats. The laboratory is equipped with Mini-2p (Thorlabs) for imaging neuronal activity in behaving animals, two 2-photon mesoscopes (Thorlabs) for imaging large cortical and subcortical areas in awake head-fixed animals, two 2-photon microscopes for single-cell electrophysiological experiments in brain slices (Chun et al, Science 2014; Blundon et. al. Science 2017; Davenport et al., Cell 2022; Bayazitov et al., Cell Reports 2024, Patton et al., Cell Reports 2024; Eom, et al, Nature Communications 2024). Preference will be given to applicants who have received their Ph.D. degrees in neuroscience or related fields within the last 3 years and have experience in imaging in vivo. Skills in computational neuroscience and data mining using machine learning methods are a plus. Qualifications: * PhD in neuroscience, or related fields * DeepLabCut or similar methods * Demonstrated hands-on experience with 2-photon imaging techniques * Experience analyzing time-series data, preferably from neuroimaging or behavioral studies * Excellent communication skills and ability to work in a collaborative environment The successful candidate will join a diverse team of researchers whose efforts are directed toward understanding behavior at the levels of neural circuits, individual neurons, synapses, and molecules.     Join Our Team The Department of Chemical Biology and Therapeutics is seeking a highly motivated and innovative scientist specialized in drug discovery to join our highly collaborative environment. The incumbent will be responsible for assay design, optimization, and validation using automation, execution of high-throughput small-molecule screening campaigns, and analysis of large, complex datasets to identify and prioritize hit compounds. Experience in both biochemical and cellular assays preferred. The candidate will work in collaboration with cross-functional teams, including biologists, computational and medicinal chemists. Projects will focus on advancing cutting-edge drug discovery programs for childhood catastrophic diseases using the latest approaches in proximity-based targeting. Specifically, based on the mechanistic basis of disease, our goal is to develop unique chemical approaches (protein degraders, synthetic gene regulators and/or functional effectors) to degrade or manipulate targets relevant to pediatric oncology, neurodegenerative disorders and sickle cell disease. Job Responsibilities: Design, develop, and optimize biochemical and cell-based high throughput screen-compatible assays for small molecule screening. Execute HTS campaigns and analyze large, complex datasets to identify and prioritize hit compounds. Collaborate with CBT Centers to progress validated hits through the drug discovery pipeline. Stay current on advancements in induced proximity and HTS methodologies, incorporating new tools and strategies where appropriate. Coordinates multiple aspects of research projects and carries out laboratory experiments utilizing various laboratory techniques, and determines whether additional or other tools or methods are required Provides input regarding research protocols and experiments. Sets up protocols and assists in developing new laboratory techniques as required. Identifies experimental data pertinent to the analysis problem. Processes, organizes and summarizes data, reporting these experimental results as required. Monitors quality and integrity of recorded information and data May draft sections of research manuscripts. Provides basic lab operation support to include equipment maintenance and purchasing. Monitors inventory levels and orders lab supplies as needed. Performs calibration of instruments and equipment. Troubleshoots instrument performance and equipment issues. Maintains regular and predictable attendance Performs other related duties as assigned in order to meet the goals of the department and institution Minimum Education and/or Training: Bachelor's degree in relevant scientific area is required. Experience in industry and a PhD highly preferred Minimum Experience: Eight (8) years of relevant work experience is required Six (6) years of relevant work experience is required with a Master's degree Three (3) years of relevant work experience is required with a PhD Prior experience in relevant lab techniques preferred. Experience in lab and scientific project coordination, maintenance, and other research support activities preferred.
