Scripps Research is seeking a Postdoctoral Associate for the Chemistry Department. The project will test the hypothesis that pharmacologic activation of the ATF6 or XBPs arm of the Unfolded Protein Response stress-responsive signaling pathway will alter the kinetic stability of transthyretin ( TTR ), possibly by altering the packing of the F87 residue. We will study whether secretory proteostasis network pathways alter the structure and modify the aggregation propensity of TTR . We will also assess whether arm-selective unfolded protein response inhibition during stress reduces TTR kinetic stability enhancing amyloidogenicity.

Research Associates are appointed positions for the purpose of developing scholarly competence, working under the supervision of a TSRI Faculty member. These appointments are normally not renewed beyond a total of six years. Research Associates cannot apply for external funding either as a Principal Investigator or Co-Principal Investigator without prior approval from a Departmental Chair. The Lotz lab in the Department of Molecular Medicine has an opening for a Postdoctoral Associate. We are looking for a highly motivated individual with strong background and technical expertise in cell and molecular biology and bioinformatics for our program on transcription factors in joint biology and arthritis pathogenesis. In this newly funded program we are using Omics technologies ( RNA -seq, single cell- RNA -seq, ChIP-seq and epigenetic analyses) to identify regulatory gene networks and test their role using in vitro and in vivo models in mutant mouse strains. Genes that are identified to be essential regulators of homeostatic processes or to promote arthritis pathogenesis are used in drug screening assays. The laboratory has preclinical models for proof of concept validation of drug targets and drug candidates.

The research project will focus on NK cell-based vaccination protocols and immunotherapies to prevent HIV infection, or attack latently HIV infected cells in models of humanized mice that harbor a functional human immune system. The work will be performed using in-house generated mice and humanized mice and requires expertise in the generation and experimental use of mice and humanized mice, cancer immunology, tissue culture, in vitro cellular assays such as quantitative PCR , killing assays, proliferation assays, flow cytometry, cellular and molecular biology. Positions require successful completion of required training courses (i.e. animal use, radioisotope training, BL-2 safety and general lab and chemical safety training).
